Research Abstract	がんによる死亡原因の4番目に多数を占める膵臓がんは、最も治療の困難ながんの一つとして知られている。その発生数は我が国においても年々増加の傾向にある。膵臓がんの治療が困難なのは、原発巣が発見されたときには既に遠隔転移が形成されつつあるためである。本研究では(1)ヒト膵臓がんのヌードマウス肝転移モデルを確立する。これに基づき、ヒト膵臓がん高転移細胞株、低転移細胞株、及びそれらのクローンを確立する(動物レベル)。(2)肝臓構成細胞(肝実質細胞、クッパー細胞、類洞内皮細胞、イトウ細胞)との共培養系などを用いて、高転移性膵臓がん細胞の相互作用する宿主細胞を同定し、高転移性細胞の転移性増殖をインビトロで測定する実験モデルを構築する(細胞レベル)。(3)高転移性細胞に高発現または欠失した分子形質を同定し、高肝転移性を規定する膵臓がん細胞の生化学的な特性を明らかにする(分子レベル)。(4)肝転移に対する治療薬の候補をこれらのアッセイ系で査定する、などである。既に前年度、ヒト膵臓がんのヌードマウス肝転移モデルを確立した。平成12年度には、臨床病理標本において転移性の高い浸潤癌と非浸潤癌のmRNA発現をDNAアレイ法で解析したところ、TIMP-1などの発現レベルに差異を見い出したので、実験モデルで高肝転移性であったヒト膵臓癌細胞株や臨床病理標本で競合的PT-PCR法で遺伝子発現レベルを比較したが、検体による差が大きく転移性との相関は見られなかった。また、Poly I/C-cationic liposome complexに対する感受性を比較し、感受性と個々の膵臓がん細胞株の特徴との間の相関に関してさらなる知見を得た。
