Research Abstract	アクチビンは、細胞増殖・分化、発癌、可塑性の制御に関わる細胞増殖因子である。本年度は、新規フォリスタチン様因子の解析を行った。また、アクチビンのシグナル伝達機構における新たな細胞内の活性制御機構の存在を明らかにした。本研究において、新規のアクチビン結合活性を有するフォリスタチン様分子(FLRG,follistatin-related gene)を同定した。この分子は、アクチビンや骨形成因子の活性を細胞外から調節する。マウスFLRGの遺伝子構造解析を行った。FLRGのプロモーター部位にはSmad結合部位、NFkappaB結合部位が存在しTGFβ/アクチビンやTNFにより活性が制御される事が示唆された。マウスとヒトのFLRGに対する非常に特異的なモノクローナル抗体の作製に成功した。遺伝子改変マウスの解析を開始した。アクチビンのタイプII受容体セリンキナーゼと会合するアダプター分子(ARIP,activin receptor interacting protein)群の解析をさらに推進させた。ARIP1は最もC末端のPDZドメインを介してアクチビン受容体と会合し、SmadとはWWドメインで会合するが、PDZドメインを介しで、癌抑制因子であるPTENやカテニンとも会合することが明らかとなった。PTENは多くのヒト癌において変異が見られるフォスファターゼである。ARIP1とPTENの会合がアポプトーシスの抑制に重要なAktの活性を制御している事が示唆された。また、ARIP群の中にARIP1とは異なった機能を有する、PDZ領域を1つだけ含有した分子が存在することが明らかとなった。
