| Research Abstract |
シトクロムP-4501A1(CYP1A1)がレチノイン酸合成酵素であることを証明しました。このレチノイン酸生合成はO^<18>を使用し、レチナールモノオキシゲナーゼであることをレチナールへのO^<18>の添加により確認しました。このP-4501A1が細胞、特に皮膚のfibroblastにとって紫外線防御作用に必須であることを発見しました。その理由は細胞内でレチノイン酸合成酵素は現在4種類の存在が確認されていますが、色素性乾皮症(XP)のfibroblast細胞は、シトクロムP-4501A1(CYP1A1)が欠損し、レチノイン酸合成活性も著しく低下していることが確認されました。in vitroの実験、特に培養細胞等ではall-trans又は9-cis-レチノイン酸添加で紫外線照射による細胞死が著しく減少することが分かりました。それ故、レチノイン酸が紫外線照射による細胞死を減少させる機構についてはいろいろ考えられますが、現在検討中です。このXP細胞が紫外線に抵抗性がない原因の1つは、このCYP1A1の欠損によります。レチノイン酸を添加すれば、紫外線保護作用が顕著です。レチノイン酸受容体については正常細胞やXPの2種のレチノイン酸受容体(RARαとRARγ)についても質的並びに量的にも解析しましたが、全く異常なく同じ量であることが確認されました。
