Research Abstract

(1) Atherectomy specimens taken from patients with unstable angina were examined by immunohistochemistry. Smooth muscle cells (SMCs) and macrophages showed positive staining for tissue factor (TF) and TF pathway inhibitor (TFPI). Staining intensity for TFPI was poor in SMCs in proliferative phase and strong in endothelial cells of microvessels. To elucidate the role of TFPI in vascular wall, TFPI gene transfer in vascular SMCs was performed by adenovirus vector. Migratory activity of TFPI overexpressed SMCs was decreased, and thrombus formation and neointimal growth were markedly inhibited in TFPI overexpressed carotid arteries. These results suggest that TFPI immunostaining of atherectomy specimens would be useful for prediction of thrombosis and/or restenosis.
(2) Cancer tissues (colon cancer and lung cancer) were also examined by immunohistochemistry. There was no correlation between cancer cell differentiation and staining intensity for TFPI.In colon cancer tissues, cancer cells in metastatic lesions were stained positive for TFPI more intensively than those is primary lesions. These results suggest that TFPI may be involved in cancer metastasis.
