| Research Abstract |
肝細胞増殖因子活性化因子(Hepatocyte Growth Factor Activator,HGFA)は肝細胞増殖因子(HGF)を特異的に活性化させるセリンプロテアーゼで、その活性調節因子であるHGF activator inhibitor type-1(HAI-1)及びtype-2(HAI-2)とともに東京工大の喜多村らのグループによって発見された蛋白である。本研究ではHGFA,HAI-1,HAI-2遺伝子欠損マウスを作製し、生体内でのこれらの蛋白の機能解析とともに、Kunitz型インヒビターのインヒビターとしての作用以外の本来の機能を明らかにすることを目的としている。
すでにマウスHGF Activator,HAI-1,HAI-2 cDNA及びGenomic DNAをクローニングしたので、今年度はクローン化したマウスHGFA,HAI-1,HAI-2 genomic DNAにmutationを加え、遺伝子を不活化したtargeting vectorを作製し、第一段階として、熊本大学動物資源センターの山田源教授らのグループの協力を得て、ES細胞にエレクトロポレーション法によりHGFAのTargeting vectorの導入を行なった。相同組み替え体をポジティブネガティブ選択法によって選別、8細胞胚に注入した後、偽妊娠マウスに移植しキメラマウスを得た。さらにC57BL/6マウスと交配しヘテロマウスを得て、十数匹のホモ欠損マウスを作成した。また、HAI-1,HAI-2遺伝子についてもgenomic DNAにmutationを加えたTargeting vectorを作成し、ES細胞へのエレクトロポレーションを熊本大学に依頼した。
