| Research Abstract |
平成12年度は以下の項目について検討した。
1.マーカー遺伝子導入微小転移モデルの作成
今年度はヒト胃癌細胞株へのGFP遺伝子導入による腹膜微少転移モデル(GCIY-EGFP)を作成し、さらに腹膜微少転移に対する抗ガン剤等の効果を青色光の外部照射により、非侵襲的かつ簡便にモニターできる新しい微少転移治療モデルを確立した。
2.微少転移形成過程のリアルタイム解析
今年度は、昨年度GFP遺伝子導入癌細胞とIntravital videomicroscopy(IVVM)法を組み合わせて確立したリアルタイム解析法および共焦点レーザー蛍光顕微鏡とラミニン螢光抗体法を組み合わせ解析システムを用いて、マウスの肝転移初期過程を解析した。ラット舌癌由来高、低転移性細胞を用いて転移過程を比較したところ、両者の転移能の違いは肝類洞内皮基底膜への安定した接着の違いに起因することが明らかとなった。
3.内皮細胞を可視化できるトランスジェニックマウスの作出に関する予備的検討
血管内皮細胞において特異的に発現しているTIE2等の遺伝子をプロモーターとし、GFPをリポーターとする発現ベクターの構築を継続して行っている.
4.胃癌患者の腹腔洗浄液中癌細胞のリアルタイムPCR法を用いた迅速定量法の意義
今年度は昨年度確立した定量的リアルタイムRT-PCR法(LightCycler)を用いて、241例の胃癌患者の腹腔洗浄液を検討した。Cut-off値を設定することにより、早期胃癌症例の偽陽性を低減させることが可能となり、診断の特異性が向上した。またCEA mRNAの定量値から腹膜再発のリスクを定量性をもって評価することが可能となった。本法は今年度厚生省から高度先進医療の認定を受け実用化の一歩を踏み出した。
