| Research Abstract |
平成13年度(最終年度)は以下の項目について検討した。
1.マーカー遺伝子導入微小転移モデルの作成
今年度は昨年度に引き続き新たにヒト胃癌細胞株(MKN28)および大腸癌細胞株(COLM-5)へのGFP遺伝子導入による腹膜およびリンパ節微少転移モデルを作成した。これらを用いて腹膜およびリンパ節微少転移に対する抗ガン剤等の効果を青色光の外部照射により、簡便かつ非侵襲的にモニターできる微少転移治療モデルを確立した。
2.微少転移形成過程のリアルタイム解析
昨年度および今年度に作成した3種類の腹膜転移性GFP遺伝子導入癌細胞を用いて腹膜転移の初期過程のin vivo解析を行った。その結果、腹膜転移には大網に限局して初期転移巣を形成する型と大網と壁側腹膜の両方に初期から転移する型の2種類およびその中間型が存在することを明らかにした。単層中皮細胞培養系を用いたin vitroの実験から両者の違いは単層中皮細胞への接着性の違いに起因する可能性を示唆した。
3.内皮細胞を可視化できるトランスジェニックマウスの作出に関する予備的検討
血管内皮細胞において特異的に発現しているTIE2等の遺伝子をプロモーターとし、GFPをリポーターとする発現ベクターの構築を継続して行っている。
4.胃癌患者の腹腔洗浄液中癌細胞のリアルタイムPCR法を用いた迅速定量法の意義
腹膜再発のリスク評価に対する定量的リアルタイムRT-PCR法を用いたPCR洗浄細胞診の意義を確認するため今年度も昨年度に引き続きProspective study(厚生労働省から認定を受けた高度先進医療として)を行い、約50症例の収集を行った。これまでのところ、細胞診に比べ感度が高く腹膜再発のハイリスク症例の選別に有用であることを示す結果を得ている。
