Research Abstract	I)RA患者単球・顆粒球におけるシクロオキシゲナーゼ2遺伝子発現の制御 1)RA患者好中球のPGE2産生が亢進しており,COX-2遺伝子発現の誘導を確認した。 COX-2活性の上昇に対するIL-4・IL-10・ステロイドの作用を検討した。 COX-2活性上昇におけるMAPキナーゼの関与を検討した。これらをまとめて論文投稿中である。 2)RA患者における末梢血単核球の活性酸素酸性に対するIL-10の制御活性をあきらかにした。 SLE患者サンプルにおける同活性を比較検討し,まとめて論文投稿中である。 II)ヒト単球のCD40発現による炎症惹起過程の機構解析 1)GM-CSF,IL-3によるCD40誘導およびCD40を介したCOX-2誘導過程について,シグナル伝達機構を中心に分子生物学的に検討した。 2)これらの誘導に対するIL-4・IL-10の作用について検討し,その機序を分子レベルで検討した。 III)IFN-γならびにIL-10のリガンド・レセプターについて遺伝的多型を発見し,その頻度を各種免疫疾患について検討した。それらを論文としてまとめて発表した。 IV)IL-4・IL-10によるシグナル伝達過程を単球系細胞について検討し論文として発表した。
