正常大腸粘膜及び潰瘍性大腸炎粘膜におけるTGFβ1産生能の部位的差異

Research Abstract	私共はこれまで大腸の近位と遠位の粘膜に於けるTGFβ1の産生量の差が大腸癌の発生頻度に影響を与える可能性について検討してきた.特にTGFβ1遺伝子のpromoter領域の%methylationは上行結腸を1とした場合、横行結腸1.4、下行結腸1.58、S状結腸1.7、直腸で2.4で、遠位大腸でメチル化の程度が強い事が明らかとなった。更に、methylation supecific PCRを用いて、その領域に含まれるCpGサイトにつき、CからTへの変換が見られない(メチル化C)クローンの陽性率を検討したが、その変換率は上行結腸で0%、以下横行、下行、S状結腸、直腸では10%、10%、15%、25%であった。これらの結果は遠位大腸では近位大腸に比して平均でTGFβ1遺伝子のメチル化の程度が高く、大腸に於けるTGFβ1の産生量の部位差にこの遺伝子のメチル化が関連する事を示すものと考えられた。また、大腸ポリープ、大腸癌近傍の非癌部粘膜におけるmethylationの頻度を正常部粘膜と比較したが、腫瘍近傍では有意にその頻度が高かった.以上の結果はTGFβ1遺伝子のmethylationによって遺伝子発現が低下し、TGFβ1の細胞増殖抑制機構に破綻をきたすことが、大腸癌発生に影響を与えている可能性を示唆するものである.
