| Research Abstract |
我々はC型肝炎の病態を規定する宿主側の因子の一つとして、レドックス(REDOX:reduction and oxidation)制御機構に着目し、C型肝炎の病態におけるレドックス制御機構の果たす役割について解明し、C型肝炎の癌化のメカニズムに迫りたいと考えている。
酸化ストレスのシグナル伝達に重要な役割を持っているシグナル伝達分子としては、NFkB(Nuclear factor kB)あるいはSTAT3(Signal transducer and activator of transcription3)が知られている。これらの分子は酸化的ストレスを受けると、リン酸化され、活性化される(NFkBの場合はinhibitorであるIkBがリン酸化を受け、分解され、活性化される)ことが知られている。そこで我々は、前年度の基礎的検討をもとに、STAT-3やIkBのリン酸化を指標として、細胞内のレドックス環境の評価を試みた。具体的には健常人およびC型慢性肝炎患者の末梢血単核球をRPMI1640(+10%FBS)培地で培養し、H_2O_2(100mM)を添加し、10-30分間培養後、細胞内蛋白を回収し、SDSポリアクリルアミドゲル電気泳動を行い、IkBおよびSTAT3に対する抗体およびリン酸化特異的抗体を用い、Western blottingによる検出を試みたが、健常人およびC型慢性肝炎患者間で有意な差は認められなかった。また、酸化的ストレスによるNO量の変動あるいは酸化(カルボニル化)蛋白質の量的変動についても検討を行ったが、健常人およびC型慢性肝炎患者間で有意な差は認められなかった。測定条件の再検討が必要と考えるがそれとともに、今後、他の指標、たとえば、酸化的ストレスの指標として最近話題になっているミトコンドリアDNAの変化についても検討したいと考えている。
