Research Abstract |
We tried to induce a T cell response to hepatitic C virus (HCV) NS5A protein by using a DNA vaccine in mice. We immunized mice with a plasmid encoding HCV NS5A region alone or with an adjuvant plasmid encoding IL-2, GM-CSF, or CD86. Neither of them induced a cytotoxic T lymphocyte (CTL) response of a siginificant magnitude. Next we induced mature dendritic cells with IL-4 and GM-CSF from bone marrow cells. We immunized mice with dencritic cells pulsed with reportedly immunogenic CTL epitopes. None of these mice showed a better T cell response yet.
