Research Abstract |
1)伴性劣性高IgM症候群(XHIM)におけるCD40リガンド異常の検討 伴性劣性高IgM症候群の責任遺伝子がCD40リガンドであることを明らかにしたが、国内症例を多数集積し、新規遺伝子異常を見出した。遺伝子診断を簡便にするためSSCP法も確立した。また、患者血清中の可溶化CD40リガンドをELISAで測定し、典型的な患者では可溶化CD40リガンドが全く検出されないことを明らかにし、血清診断が可能であることを示した。また、保因者では、可溶化CD40リガンドが低下していることも見出した。 2)CD40シグナル伝達異常による高IgM症候群の病態解析 女性の高IgM症候群患者を見出し、その末梢血B細胞を可溶化CD40リガンドで刺激し、増殖、表面マーカー発現、IgEクラススイッチの異常の有無を検討した結果、増殖が障害されている症例、表面マーカー発現が障害されている症例、IgEクラススイッチが障害されている症例、など、多彩な異常が見出された。 3)CD40シグナルに関わる分子の検討 CD40シグナルに関わる分子として、Kuを同定した。すなわちKu分子が、CD40細胞内領域に結合し、刺激後チロシンリン酸化し、核内に移行することを見出した。また、刺激により、Ku分子を介してB細胞のDNA-PK活性が上昇することも見出した。このことは、Ku分子がCD40によるクラススイッチに関与していることを示している。
