Research Abstract |
The gene encoding 25-hydroxy vitamin D-1alpha-hydroxylase has been cloned and this allows the study on the expression of 1alpha-hydroxylase in kidney. However, there are only a few studies on its promoter activity. We have cloned the upstream region of the human 1alpha-hydroxylase gene using the BAC library and examined the promoter activity of several fragments of the region with various length. Then, we found that the 1.5 kbp region upstream the initial point of the transcription has a promoter activity. The promoter activity was high When the region was introduced into cells derived from renal tubular cells. The addition of 10^<-8> M1,25-dihydroxyl vitamin D3 suppressed the promoter activity by 25-50%, but the critical region responsible for the suppressive effect of 1,25-dihydroxyl vitamin D3 because the suppression was observed even in the shortest fragment. Next, the expression of the 1alpha-hydroxylase gene was examined in association with kidney development using 6 4 mouse kidney organ culture. 10^<-6> M-10^<-4> M forskolin induced the expression of the 1alpha-hydroxylase gene in kidney taken from 15.5 dpc mouse fetus 3 to 24 hour after the addition. The expression was observed in kidney taken from 13.5 dpc mouse fetus. Thus, the expression of the 1alpha-hydroxylase gene starts earlier than previously thought.
