| Research Abstract |
研究経過
1)担癌患者の周術期のhumonalな環境の基礎データとしてCOX-2の影響下にある血管新生因子VEGFとその抑制因子endostatinを評価した.方法として胃癌の術前患者の血漿中よりELISA法にて両者を測定し(n=35),その進行度,臨床病理学的所見や腫瘍マーカーとの関連を分析した.その結果血漿中のVEGF,endostatin共stageに比例した上昇はなく特に後者は逆相関した.すなわちこれらの因子が全身的に癌の進展に関与する可能性は否定的と考えられた.より局所的な要因を確認するために現在,腫瘍中の各濃度を測定中である.
2)in vitroの実験では胃癌細胞株や腹水癌細胞におけるCOX-2の発現とCOX-2inhibitor-JTE-522の細胞増殖とサイトカイン産生に及ぼす作用を検討した.RT-PCRではSTKM-1,MKN45とKATO3にCOX-2のは発現を認めたが,STKM-2とMKN28では認めなかった.JTE-522を添加した培養では細胞増殖に影響はなかったが,COX-2発現株ではDNAladder(apoptosis)が一部認められた.
3)in vivoではICR-nudeマウスとCOX-2を強発現しているMKN-45P胃癌細胞株を用いた腹膜播種転移モデル作成を作製し実験を行った.癌細胞を腹腔へ移植後にまず,JTE-522(10mg/kg)にて治療を開始(day1-14),その後1週間以上間隔をあけてTNP-470(20mg/kg,day14-28)を投与した.
今回の検討では腫瘍細胞の量を変えて実験を繰り返したものの(各n=36,n=30)JTE-522による有意な延命効果はなかったが,JTE群でのみ生存例を認めた(約20%).
