| Research Abstract |
大腸癌同所移植モデルにおけるin vivo治療実験(岩橋 誠・山上裕機)CEA promoterの制御下にCre recombinaseを発現するAxCEANCreとCD遺伝子を発現するAxCEACD, Creの存在下でCAG promoterの制御下にCD遺伝子を発現するAxCALNLCD, CAG promoter下にCD遺伝子を発現するAxCACDをCOS-TPC法にて作製した.ヌードマウスの虫垂にCEA産生大腸癌細胞株LoVoの腫瘍塊(2mm角)を移植した大腸癌モデルを作成し,その7日後,以下の4群(各群n=5)にadenovirus vector(3.0×10^9pfu)を腹腔内投与し,その翌日より5-FCを500mg/kg、10日間腹腔内投与した.大腸癌移植より5週後に屠殺し腫瘍重量(mg)を測定した結果,A群:MockとB群:AxCEACDでは,それぞれ251±92,215±45であるのに対し,C群:AxCEACre+AxCALNLCDとD群:AxCACDではそれぞれ46±9,68±16と腫瘍増殖の著しい抑制効果があり,A, B群と比較して有意差を認めた(P<0.001).さらに,A, B群の肝臓には5匹中それぞれ3匹,2匹に癌転移巣を認めたが,C, D群には肝転移を肉眼的にまったく認めなかった.なお,D群は腫瘍増殖抑制効果の点でC群と同等であったが,屠殺時のGOT, GPTは他群と比較し著しく上昇しており,この群のマウス5匹中1匹が早期死亡したことから,D群において強い副作用が出現することが判明した.上記の治療プロトコールで各群n=7として,生存期間を検討した結果,C群:AxCEACre+AxCALNLCDは139±12日であり,他の全ての群より著しく延長した(P<0.01).
