Research Abstract

The purpose of this study was to evaluate the action of estrogen as a vasodilator or modulator on the vascular tonus during chronic treatment with the hormone. In this study, experiments were conducted on isolated basilar artery from two groups of female rabbits : ovariectomized with no estrogen replacement (OVX) and ovariectomized with estrogen replacement (OVX+E). Arteries from OVX and OVX+E groups were contracted in 30 mM K^+ and relaxed by application of both acetylcholine and carbachol. OVX+E arteries were relaxed more strongly than OVX arteries. In both endothelium-intact and -denuded preparations, the vasodilatation induced by NOR1 or SNAP was greater in OVX+E than that in OVX arteries. Whereas the vasodilatation induced by nicardipine, 17β-estradiol and membrane-permeable cyclic-GMP or cyclic-AMP was the same in the two groups. These results suggest that chronic administration of estradiol potentiates reactivity to NO in smooth muscle cells. Modulation at the genetic level of could prove important in this effect.
