| Research Abstract |
細胞外マトリックス(ECM)や細胞膜に存在するグリコサミノグリカン(GAG)が細胞の増殖因子・成長因子や様々な活性化因子と選択的な親和性を有することが,相次いで明らかとなり,これまで単にECMあるいは細胞膜構成成分としか捉えられていなかったGAGの役割が脚光を浴びるに至っている.
申請者らが見出した新しいヘパラン硫酸素グリコサミノグリカンの生理活性の一つとして,塩基性線維芽細胞増殖因子との相互作用を明らかにした.さらにこの新しいGAG鎖が,増殖因子受容体とヘパラン硫酸の相互作用を抑制する効果について,定量的に追求した.また核磁気共鳴スペクトル法による二分子間の相互作用を定量的に解析する手法も確立した.これらの結果を基に,細胞内極微量のGAGの挙動解析を追求している.また,化学的に脱硫酸化,あるいは過硫酸化したグリコサミノグリカンを調製し,抗ウイルス活性,抗血液凝固活性あるいは細胞増殖阻止活性などを調べた結果,非常に強い表記活性を示した.現在,これらの作用メカニズムについても検討を進めている.
