| Research Abstract |
内在性μ-オピオイド受容体作動薬(endomorphin-1および-2)の学習記憶障害誘発作用におけるドパミン受容体サブタイプの関与について明確にした.
1.Endomorphin-1(10μg)およびendomorphin-2(10μg)によって受動的回避学習は有意に障害された。ドパミンD1受容体作動薬R(+)-SKF38393(0.05mg/kg)は,endomorphin-1(10μg)による受動的回避学習障害を改善する傾向を示したが,R(+)-SKF38393(0.1mg/kg)あるいはドパミンD1受容体拮抗薬R(+)-SCH23390(2.5および5μg/kg)は,endomorphin-1(10μg)およびendomorphin-2(10μg)による受動的回避学習障害にほとんど影響しなかった.
2.ドパミンD2受容体作動薬RU24213(0.3mg/kg)は,endomorphin-2(10μg)による有意な効果を消失させたが,RU24213(1mg/kg)はendomorphin-1(10μg)およびendomorphin-2(10μg)による受動的回避学習障害にほとんど影響しなかった.ドパミンD2受容体拮抗薬(-)-sulpiride(10mg/kg)は,endomorphin-1(10μg)ではなく,endomorphin-2(10μg)による受動的回避学習障害を抑制した.
3.ドパミン受容体作動薬・拮抗薬単独は,受動的回避学習にほとんど影響しなかった.
以上の結果より,ドパミンD2受容体を刺激すると,endomorphin-1ではなく,endomorphin-2による受動的回避学習障害が惹起されることが示唆された.
