HGFクリングルによる血管抑制と新規クリングル分子の同定・血管調節に関する研究

Research Abstract

血管新生は組織の再生において必須の機能を担う一方、病態の進展にも関与する。とりわけ、血管新生促進は心筋梗塞、閉塞性動脈硬化症などの疾患治療に応用されうる一方、血管新生阻害因子は腫瘍血管新生阻止に基づく新しい制癌剤のターゲットとして広く注目されている。本年度においては、肝臓をはじめとする各種臓器の再生・保護において中心的役割を担うHGFの分子内断片(NK4)が強力な血管新生阻害作用を発揮するとともに、新しい制癌剤となる可能性を見いだした。HGFは4個のクリングルドメインを含むα鎖とセリンプロテアーゼ様ドメインをもつβ鎖からなるヘテロダイマー蛋白質であるが、エラスターゼによるHGFの消化産物から、4個のクリングルドメインを有する分子内断片(NK4)を得た。興味深いことに、HGFは強力な血管新生因子の一つであり、血管内皮細胞の増殖や遊走を強く促進したのに対して、その分子内断片であるNK4はHGFに加え、bFGF、VEGFといった代表的な血管新生因子によって促進される血管内皮細胞の増殖ならびに遊走をほぼ完全に阻害した。さらに、NK4はin viroにおける代表的血管新生のアッセイ系であるウサギ角膜法、ニワトリ奬尿法膜法においても血管新生を強力に阻害した。これらの結果に基づき、NK4が腫瘍血管新生阻止に基づく新しい制癌剤になる可能性を検討した。ヌードマウス皮下にヒト胆嚢癌、マウス肺癌、ヒト乳癌を移植し、これらマウスにNK4を投与した。その結果、NK4を投与したマウスでは、いずれの腫瘍組織においても、腫瘍血管新生が阻害され、これにより癌の成長は強く抑制された。また、NK4はこれら癌の肺への遠隔転移を強力に阻害した。これらの結果はNK4が腫瘍血管新生阻止に基づく新しい制癌剤になる可能性を示している。本年度においては、上記NK4の研究とともに、クリングルドメインを有する分子が血管組織において重要な機能をもつことから、クリングルドメインを有する新規分子のクローニングを行った。各種クリングルドメインにおける高度保存領域とそれぞれのクリングルに特有の可変領域の塩基配列を利用して、クリングルドメインをコードするcDNAを一挙に解析する新規方法を確立し、この方法によって新規クリングル分子(MBK-1)をクローニングした。この新規クリングル分子は細胞外にクリングルドメインならびにCUBドメインといった特徴的構造をもつ膜蛋白質であり、その構造から、本分子はレセプター様分子として細胞間相互作用をメディエートすることが予想され、現在その機能解析を勧めている。
