Research Abstract

1)滑膜肉腫にみられる染色体転座t(x;18)(p11.2;q11.2)より生じたSYT-SSX遺伝子産物の抗原ペプチドPYGYDQIMPK、YGYDQIMPKKがHLA-A24拘束性のキラーT細胞(CTL)エピトープになることが明らかにされ、この腫瘍の癌ワクチンの候補となることが示された。
2)網膜と癌組織に発現するrecoverin蛋白質が癌患者にみるcancer-associated retinopathy(CAR)の原因抗原蛋白と考えられ、特にHLA-A24拘束性のR49ペプチド(QFASIYAKF)などrecoverin由来の3つのペプチドがCTLの標的となることが示された。癌ワクチンとしてばかりでなく、自己疾病発症機序の研究としても特にCARは大きな意味をもつものと考えられた。
3)口腔癌OSC-20でCD4T細胞の標的抗原がHLA-DR8拘束性αエノラーゼ由来の16アミノ酸よりなる抗原ペプチドであることが示唆された。
4)ヒト肺癌(LHK-2)、膵癌(PUN)、骨肉腫(OS2000)、咽頚ヘン平上皮癌(STK-1)などで自己CTLクローンを得た。現在抗原遺伝子クローニング中である。
