Research Abstract	現在までに、クローン病炎症腸管粘膜においてインターロイキン18(IL-18)の発現が浸潤マクロファージにおいてIL-12とともに増強し、腸管粘膜内リンパ球の増殖因子として機能を有することを報告した(Kanai T et al.Gastroenterology.2000 in press)。我々は、今回、慢性腸炎におけるIL-18の関与を直接的に証明する目的にて、遺伝子工学的に作製した慢性腸炎モデルマウス、TCRα鎖ノック・アウト(KO)マウス、IL-7トランスジェニック(Tg)マウスを用いて、IL-18/IL/18R機構の解析を行った。(成績)1)TCRα鎖KOマウス、IL-7・Tgマウス腸管粘膜内リンパ球のIL-18R発現の増強、粘膜内マクロファージのIL-18の発現の増強を認めた。2)TCRα鎖KOマウス、IL-7・Tgマウス腸管粘膜内リンパ球において、IL-12、IL-18は増殖因子として機能することを示した。3)TCRα鎖KOマウスにおいて、抗マウスIL-18抗体によって発症が抑制されること、IL-18の活性化因子であるICE分子が関与することを証明した。次年度はTCRα鎖KOマウス、IL-7・TgマウスにおけるCryotopatch構造の免疫染色での比較検討、また、同構造における腸炎発症時のIL-18/IL-18R機構の解析、実体顕微鏡下に分離したリンパ球の細胞表面マーカー、サイトカイン産生能を評価し、腸炎発症の機序を追究したい。また、分離した未分化リンパ球の培養を行い、分化発達におけるIL-18、IL-7の関与につき検討する。
