胚・子宮内膜細胞相互作用の基礎の解析と胚移植法への応用

Research Abstract

我々はマウスにおいて胚-子宮内膜間質細胞共培養系を確立し、胚による子宮内膜間質細胞分化制御機構、および子宮内膜間質細胞脱落膜化による胚発育促進機構の存在を明らかにし、マウス初期胚により子宮内膜間質細胞に誘導される遺伝子を同定し、d/tPRPの他decidual PRLがあることがわかった。マウスにdecidual PRLが存在することを初めて証明し、さらにこの誘導は初期胚から分泌される、プロゲステロン以外の液性因子によることも明らかにした(投稿中)。現在これらの因子について実体を明らかにしつつある。また、マウスにおいて胚移植法を用いた着床成績の解析で、胚の存在により、子宮内膜の胚受容能が向上し、子宮内膜間質細胞のPCNA発現が亢進することを明らかにしているが、マウス子宮内膜内胚移植をおこない、その凍結切片をもちいてマウスProlactin、IGFBPsなどについて胚移植前後でin situ hybridizationをおこなった。以下のように一部は既に学会発表した。1)竹林浩一、高倉賢二、木村文則、野田洋一;ゴナドトロピン異常を伴う排卵障害におけるゴナドトロピン受容体の遺伝子解析の試み;第52回日本産科婦人科学会学術集会・2000年4月徳島市2)木村文則、高倉賢二、竹林浩一、笠原恭子、後藤栄、中西桂子、野田洋一:胚依存性子宮内膜分化機構存在の証明-胚による子宮内膜プロラクチン遺伝子の誘導;第52回日本産科婦人科学会学術集会・2000年4月徳島市3)Kasahara K,Takakura K,Takebayashi K,et al.;Different response of cultured human endometrial stromal cells to five progestogens used for luteal support.International Symposium on Cell and Molecular Biology of Endometrium in Health and Disease,October3-5,2000,Kobe4)Kimura F,Takakura K,Takebayashi K,et al.;Embryo-dependent gene expression in mouse endometrial stromal cell(ESC)in vitro.International Symposium on Cell and Molecular Biology of Endometrium in Health and Disease,October3-5,2000,Kobe5)Takebayashi,K.,et al The follicle-stimulating hormone receptor gene mutations are rare in Japanese women with premature ovarian failure or polycystic ovary syndrome.56Annual Meeting of the American Society for Reproductive Medicine.October21-26,2000,San Diegoこれらの基礎的研究をもとにわれわれは新しい胚移植法を開発した(投稿中)。
