| Research Abstract |
癌に対するバイオセンサー型DNAチップを作製するため、以下の研究を行った。
1)DNAチップとの反応・測定条件の設定
1000ほどの遺伝子を用いて、ガラス基盤上にDNAチップを作製し、ヒト肝細胞癌由来細胞株を用いてhybridization,washing、検出等の条件設定を行った。また細胞や組織からのRNA抽出に加えて、微量サンプルを扱うためにRNA amplificationの条件設定を行い、mgオーダーの組織から安定してDNAチップに持ち込む系を確立した。またいくつかのコントロール条件を設定した。
2)遺伝子情報解析の構築
得られた遺伝子情報は膨大なものとなるため、遺伝子情報および解析情報をソフトウェアを用いて入力し、インターネット上の情報とも連動して解析するながれを構築した。また疾患に対して得られる遺伝子情報にたいして、疾病との連動を検討する、あらたな解析ソフトの開発を開始した。
3)バイオセンサー型チップ作製条件の設定
従来のDNAチップ作製装置を改良し、基盤上の微小な金属電極にDNAをスポットする装置を開発した。コンピューター上で、異なる微小電極の配置に対しても対応できるソフトウェアを開発している。
4)バイオセンサー型チップの反応・測定条件の設定
基盤上にリソグラフィーの原理を応用し、微小電極を作製した。この電極上にDNAを載せ、検体DNAとの反応条件、インターカレ一夕ーとの反応条件、測定条件等を検討した。用いたウイルスDNAに対して微小電流が得られ、その測定条件の設定を開始した。
