| Research Abstract |
肺癌切除例は,たとえI期,II期であっても遠隔転移を中心とした再発死亡が少なくないが,現在のところ,肺癌の術後化学療法の延命効果は明らかでないと言わざるを得ない.TNP-470などの新しい血管新生抑制剤が臨床治験に入ったが,副作用も多く,実際に本邦においてAdjuvant therapyとして使用できるまでには相当の年月が必要と思われる.一方,日常診療で用いられている薬剤のうちのいくつかは,強力な血管新生抑制作用を持っている.その中でもマレイン酸イルソグラジン,トラニラストなどの血管新生抑制作用は強力であることから,これらの薬剤の血管新生抑制作用を種々の点から検討した.
Bovine capillary endotherial cell/Human umbrical vein endotherial cellをI型コラーゲンゲル上でbFGFと添加薬剤の存在下で培養し,ゲル内に侵入して形成した管腔の長さを定量的に解析し,上記の2薬剤とも血管新生抑制作用を示した.また,B57マウス皮下にbFGFを含むMatrigelを移植した後,薬剤を経口投与し,Matrigel内に浸潤した内皮細胞を計測し定量化することにより,薬剤の効果を検討し,やはり血管新生抑制作用を有することを示した.
