Research Abstract	法医学において個人識別は極めて重要な研究分野の一つであるが、現在のところ血液などの体液から年齢推定可能な年齢依存性変化を示すマーカーはなく、これらマーカの発見・法医学実務への導入が図られなければならない。そこで、本研究では血液や体液から年齢推定に活用しうるマーカーを開発すべく予備実験、文献調査および討議を重ね、本課題を将来「重点領域研究」の研究課題とする予備調査を目的とした。 1.従前の研究において17歳以下の若年者尿中には存在するが、それ以上になると検出できなくなる年齢依存性タンパク質(Ugl-Y)を見出した。今回、若年者尿から精製したUgl-Yに関するタンパク質化学的解析から、Ugl-Yの全アミノ酸配列の95%以上を決定した。さらに、その構造はfibronectinの一領域にほぼ一致することが明らかとなった。2.年齢推定に利用しうる年齢依存性生体分子を検索するため分子生物学的アプローチとして遺伝子発現パターンの年齢依存性変動を精査した。遺伝子発現の変動をモニターする有効な方法であるfluorescence differential display(FDD)-PCR法を利用して、異なった成長段階におけるマウス腎臓の遺伝子発現を解析した。その結果、7種類の遺伝子で有意な年齢依存的発現の認められることを見出した。そのなかには老年期のみに発現されるものなど様々な年齢依存的発現パターンが観察された。本研究結果はFDD-PCR法が年齢依存的発現を示す遺伝子の検索には有効な手段であることを証明するものであった。今後は、本法によって見出された年齢依存的発現を示す遺伝子について、その産物が法医学的試料から検出しうるものか否かを検討して年齢推定マーカーとして活用する予定である。以上の研究成果、文献調査などから、年齢依存性マーカーの発見・実務への応用に関する基本的コンセンサスが得られ、それに沿って研究を継続する予定である。
