Research Abstract	各班で目的にそって以下の成果をあげた。・人工複製系構築班:試験管内での複製系構築は、人工生命への一歩につながるもので、それ自体意義があるが、さらに、再帰的な増殖での少数の遺伝子の意義を明らかにできた。一方で複製系では少数分子が他をコントロールするようになり、情報を担うようになるという理論を発表した。このように、理論と実験が対応する結果をしめせた。これは生命が何故遺伝情報を有するように至ったかに迫る結果である。ついでリポソーム内でDNA->mRNA->たんぱく質の反応系を構築し、さらに、これを淘汰させていくことで複製能力を高めるような進化がおこることを示した。一方で、養分を内包したジャイアントリポソームの自己生産に成功し、再帰的増殖系の実験にあと一歩までこぎつけた。これと関係して、2種の両親媒性分子を混合した系で分裂を誘発していくダイナミクスを見出した。このリポソーム増殖系とRNA複製系を組み合わせて、「内部で反応をしながら増殖していく膜」としての細胞プロトタイプを作る準備を進めている。また、理論としては膜、細胞内反応の増殖系がいかにたがいに同期して再帰的増殖系を構成できるか、その際の反応ネットワークの特質は何か、またそこで遺伝情報を担う部分が現れるかが重要課題でありそれに向けてのモデル数値計算が進行中である。・発生過程制御班:既に大腸菌の相互作用による細胞分化はえられてきたが、大腸菌内に細胞分化の指標となるミニネットワークを持った遺伝系構築ができ、分化過程の定量的追跡が可能になり、大腸菌集団が相互作用によっていかに状態を分化させるかの研究準備は完成した。また、これをふまえて「多細胞生物のプロトタイプ」をつくる実験も、相互作用の効果が実験的に明らかになり着実に進んでいる。一方、アニマルキャップの未分化細胞からの、アクチビン、レチノイン酸、con-Aの濃度とそのタイミングの違いによる組織構築はほぼ成功した。特に、このプロジェクトで重要な点は、細胞数を制御パラメータとして各臓器形成の頻度が変化するかが明らかになったことであり、これは細胞集団全体の各細胞への影響という複雑系としての性質を抽出する重要な成果である。また構築した発生過程では、正常発生過程の中間ステップをとびこして最終状態にいくというjump-over現象が明らかにされた。これは、発生をダイナミクスを通した安定状態への吸引としてとらえる理論の正当性を示唆するものであり、理論と実験を結ぶ成果である。また、アクチビンを処理した後、5時間おいてからレチノイン酸を処理すると膵臓が形成されるというように、時間差処理によって新しい臓器形成を可能にした。これは細胞の内部に操作による変化が記憶されていることを示唆し、細胞内部状態の履歴の理論と関係している。更に、眼球の組織をアクチビン処理で構築した。しかも、眼を摘出したオタマジャクシにこの作った眼球を入れると、視覚機能を持つようになることまで確認した。一方、理論ではこれまでに細胞内部のダイナミクスの不安定性と細胞間相互作用だけを仮定して細胞分化と形態形成が、力学系モデルで一般におこることを示したが、これを進めて、幹細胞からの分化の性質、また細胞状態の可塑性と分化の関係をあきらかにしてきた。こうした実験と理論の関係を明確につけていくには細胞の性質をセルソーターで分離し、細胞の性質の分布の変化を定量的に追う必要があり、そのために新たなオンチップセルソーターの開発を進めて、ほぼ実用段階にまでなって来た。相互作用による表現型の分化が遺伝型の進化を促すという種分化の理論を昨年度発表したが、これを進めて、その条件、またその種分化の固定化を明らかにした。これは、交配でのえりごのみ過程を前提とせずに、安定した同所的種分化をおこすはじめての一般的理論であり、次第に進化研究者にも広まっている。今後この理論の適用範囲を広げて行く研究を進めて行こうとしている。特にこの理論の実験室での検証は、大きなブレイクスルーになるであろうから大腸菌人工進化系での研究を進めている。既に細胞間相互作用により進化が加速して、多様な大腸菌が共存することを検証して来た。一方で粘菌と大腸菌の共生関係形成に際しても、表現型の可塑性を確認した。ここからも理論一実験の共同による展開が進行中である。・動的生命解析班:マイクロ加工技術と光ピンセット操作を応用した1細胞長期培養観測装置系の開発、各細胞の性質をこちらで判定しながら、ダメージをあたえずに分離し培養、解析するオンチップセルソーターともに順調に進んでいる。またこれらの技術を大腸菌に用いて姉妹細胞で分裂能がどう変わるかを明らかにした。これらの結果は、論文で発表する一方、複雑系生物学の著書も上梓し、分野の浸透をめざしている。
