| Research Abstract |
ゲノム刷込み(インプリンティング)を受ける哺乳類遺伝子がゲノムの特定の場所にクラスターを形成することは、これが染色体の機能ドメインを解析するよいモデルとなることを示唆している。マウス第7染色体遠位部(7F4/F5)の刷込み領域は、糖尿病、Beckwith-Wiedemann症候群、Wilms腫瘍などの刷込み関連疾患の責任領域であるヒト第11染色体短腕(11p15.5)と相同である。ヒトの刷込み関連疾患の解析を支援するため、本年度は、このマウスの刷込みドメイン(およそ1Mbに10個余のインプリンティング遺伝子がある)の全塩基配列を佐賀医大向井ら、理研服部らと共同で決定した。ヒトの当該領域と比較した結果、例えばKvLQT1のイントロン内に進化的に保存された配列があり、それらが制御配列である可能性が示された。さらに、H19周辺の保存された配列を実際にレポーター測定法やゲルシフト法により調べ、組織特異的エンハンサーや刷込みに関わるインスレーター蛋白質の結合配列であることを報告した。また、このドメインの配列と一致するESTをBLASTを用いて検索したり、エクソン予測ソフトを行い、新規遺伝子の可能性のある配列を多数見つけた。しかし、これらの転写物の詳細な解析はこれからである。今後、同定された遺伝子や調節配列をマウス個体でノックアウトするなどにより、刷込みの制御機構やヒト刷込み疾患の発症機構を明らかにしていく予定である。
