| Research Abstract |
SR-BIはHDL受容体として生理的に機能していることが明らかになった唯一の受容体である。SR-BIはHDLが結合するとHDL中のコレステロールエステル(CE)のみを選択的に細胞内に取り込むが、形質膜の内側に取り込まれたCEが如何にして更に細胞内へと輸送・代謝されるか、その分子機構については殆ど解明されていない。我々はSR-BIのC末端細胞質ドメインに結合する蛋白質を初めて同定することに成功し、CLAMPと命名した。CLAMPはPDZドメインを分子内に4個持ち、最もN末側のPDZドメイン(PDZ1)を介してSR-BIと結合することにより、SR-BI蛋白質の発現レベルを上昇させるとともに、受容体を介して細胞内に取り込まれたCEの代謝・輸送経路を調節している可能性が細胞レベルの解析から示唆された。また、PDZ3及びPDZ4を欠失した変異型CLAMP遺伝子をアデノウィルスベクターに組み込み、SR-BI及びCLAMPの両発現細胞に感染させたところ、両者の結合が抑制されることがわかった。そこで変異型CLAMP蛋白質をマウス肝臓に過剰発現させたところ、SR-BIの発現量が減少し、血清HDLコレステロールレベルが有意に上昇していることがわかった。このことから、CLAMPは個体レベルにおいてもSR-BIを介するHDLからのCE取り込みに重要な役割を果たしていることが示され、血清HDLコレステロールレベルを調節する新しい分子機構の存在が明らかとなった。
