Research Abstract	トレニア(Torenia fournieri)の胚嚢が裸出している。この性質を利用して(1)高等植物の花粉管の胚嚢への誘導、(2)重複受精における精細胞や卵細胞及び中央細胞の受精の瞬間、(3)卵細胞内での物質の偏在と初期胚分化との関連を明らかにするとともに、(4)初期胚の形成などの分子機構の研究をおこなった。その代表的な成果について述べる。まず重複受精の最初の段階である花粉管誘導機構を解析した。受精前における胚嚢内の二つの助細胞、卵細胞、中央細胞を、紫外線レーザーで破壊し、花粉管の誘導率を調べた。その結果、受精直前の助細胞を破壊した時のみ、花粉管誘導の著しい低下がみられた。助細胞は他の細胞に比べて大量のERとリボソームを含んでいたことから、受精に先だって助細胞より選択的に受精誘導物質が放出されていることが考えられた。そこで、種々の方法で調べた結果、それがタンパク質であることが示唆され、そのタンパク質を同定するために、胚嚢細胞をプロトプラストにし、助細胞を顕微操作により集める方法の開発を行った。その結果、数10個の助細胞を集めることができた。更に規模を拡大し、受精直前に合成されるタンパク質とRNAの同定への道を拓いた。また胎座の胚発生における重要性が明らかとなり、胚発生の特定な時期に胎座から特別な物質の供給があってはじめて正常な胚発生が行われることがわかった。
