Research Abstract

今年度は、デュアルスペシフィシティホスファターゼVHRの分子構造(すでにX線結晶解析の報告有り)に基づいて、選択的阻害剤の設計、合成、作用解析を行った。VHRの活性中心には、他のチロシンホスファターゼにも保存されているCysで始まりArgで終わるループ構造(CX5R)が存在し、このループの内側を向いたアミドのNHと基質のホスホチロシンのリン酸部分が相互作用すると考えられる。さらにそのループの隣には、TyrやLeuといった疎水性アミノ酸がならぶ疎水性領域が存在する。我々が放線菌から見出し,たVHR阻害剤RK-682(テトロン酸とアルキル鎖よりなる)とVHRをコンピューター上でドッキングさせると、テトロン酸が基質のリン酸基をミミックしVHRのCX5R領域と相互作用し、アルキル鎖がVHRの疎水性領域に嵌り込むことによってホスファターゼ活性を阻害することが示唆された。一方、RK-682のVHRに対する阻害のキネティックスを詳細に検討した結果、RK-682一分子ではVHRに対し安定に結合できず、二分子必要であることが示唆された。そこで得られた結合モデルに基づいて、RK-682が二量体化した化合物RS-39Sを合成したところ、RK-682よりも約10倍強力(IC_<50>=1.8μM)で一分子結合で阻害活性を示すことが明らかになった。
