G1期におけるDNA複製因子の発現と活性の制御機構の研究

Research Abstract

増殖刺激からDNA複製開始に至るG1期制御を解析し、以下を明らかにした。1)E2Fの新規標的として、28個の既知遺伝子と8個の機能未知遺伝子を同定した。8個の機能未知遺伝子のうち5個の遺伝子の発現がDNA複製開始に必要であった。2)E2FによるARF遺伝子の特異な発現制御機序を解析し、サイクリン依存性キナーゼ(Cdk)によるリン酸化を介さずにpRBが不活性化されるとE2FがARF遺伝子の非典型的なE2F結合配列に結合し遺伝子発現を活性化することを明らかにした。3)テロメアの正常な機能の維持に重要なテロメラーゼ遺伝子発現が、ヒトT細胞においてG1/S移行期に活性化されることを見出した。その制御に重要なエレメントとそこに特異的に結合する細胞性因子を同定した。4)膵臓癌と肺癌においてJab1の発現が亢進し、それに伴ってCdk inhibitor p27の発現の低下や癌が悪性化している所見を得た。5)Jab1遺伝子座に変異を導入したマウスを作製し解析を行った結果、Jab1ノックアウトマウスは胎生致死で、cyclin E, tumor suppressor p53, Cdk inhibitor p27の発現が向上し、細胞増殖が抑えられ、アポトーシスが亢進すること、Jab1ヘテロ細胞はp27の発現上昇を伴ってS期への進行が遅くなることを見いだした。6)慢性骨髄性白血病(CML)において、原因遺伝子産物Bcr-Ablの下流でJab1が働きp27分解を促進することを見いだした。この反応経路ではRas/MAPKおよびPI3Kシグナル伝達系を介しており、CML発症に繋がる細胞増殖の一因となることを明らかにした。7)マウス線維芽細胞でJab1が細胞接着かつ細胞周期依存的に制御されること、さらに、rasによりトランスフォームした細胞ではこの制御がなくなっていることを見いだした。
