| Research Abstract |
1)チャネル蛋白質の立体構造モデルの作成とそのモデルに基づく実験結果の解析
K^+チャネルの薬理特性の研究が行われているKir6.2,Kir6.1およびATP感受性K^+チャネルのスルホニル尿素受容体の立体構造モデルの構築を行い、それぞれに表れる伝導性や薬理特性の違いを、立体構造モデルによって説明した。
2)蛋白質の表面形状と物性データベース(eF-site)と表面構造の認識・分類
蛋白質の機能発現と立体構造との関連を、分子表面形状と物性から分類・整理するためチャネル蛋白質を含むデータベースを構築・公開し、分子表面の高速識別手法としてグラフ理論を基にしたクリーク探索手法を開発した。
3)NMRのSaturation Transterを利用したチャネル蛋白質とリガンドとの複合体立体構造モデリング手法の開発
イオンチャネルとその阻害剤のSaturation Transfer実験結果(東大薬・嶋田一夫教授との共同研究)を利用し、阻害剤側の結合部位に対する実験情報を満たすように複合体モデルを構築するドッキング・シミュレーション手法を開発した。
4)シミュレーション計算による分子認識機構の解析
構造サンプリングによる演繹的モデリング手法の開発を進め、ペプチドやループ構造のモデリングと自由エネルギー地形の解析を行った。
