物質のベクトル運送を制御するCa^<2+>シグナル調節チャネル群の分子生理学的解析

Research Abstract

我々は、ショウジョウバエ遺伝子transient receptor potential(TRP)とその動物ホモログ運伝子産物が、様々の細胞刺激により活性化するカチオンチャネルを形成していうことを明らかにしてきた。本年度は、TRPC5が一酸化窒素(NO)を感知し活性化開口し、Ca^<2+>を流入させることを見出した。また、TRPC5は、血管内皮細胞の形質膜陥入構造カベオラにおいて、内皮型の一酸化窒素産生酵素eNOSと機能的複合体を形成し、内皮細胞におけるNO産生のpositive feedback loopを制御することがわかった。最近、多発性嚢胞腎原因遺伝子TRPPがTRPC3チャネル膜移行に重要な役割を果たすことも明らかにした.
小胞体はCa^<2+>取り込み活性、Ca^<2+>貯蔵活性とCa^<2+>放出活性を内在し、細胞内Ca^<2+>ストアとして機能する。筋細胞小胞体において、Ca^<2+>千取り込みとCa^<2+>放出はそれぞれ横経部(longitudinal region)と終末部(terminal region)が担当しており、Ca^<2+>の細胞内ベクトル輸送が存在する。興奮性細胞の小飽体のCa^<2+>ベクトル輸送の理解に向けて、その機構形成に寄与する分子の同定とその役割を明らかにした。ミツグミン29と命名した細胞膜タンパク質は小胞体Ca^<2+>放出やストア依存性Ca^<2+>流入の調節に重要であることをみいだした。また、筋小胞体腔タンパク質であるサルカルメニンは筋小胞体のCa^<2+>貯蔵機能に寄与していることなどを解明した。
