| Research Abstract |
本研究計画では、アジアの中の日本という立場からアジアにおけるヒトゲノム多様性計画に資する「細胞・遺伝子レポジトリー」の確立を目指している。
そこで、先ず、一般に汎用されているEBウイルスを用いた細胞株化をおこない、細胞とDNAのバンク化を開始する。次に、「安くて易い」汎用型細胞株化法の開発をおこなう。本研究の緊急性と現地の対応に鑑み、ヒトゲノム多様性ワークショップでノミネートされたアジア・オセアニアの稀少集団も対象とするが、各地域の主要な集団を対象にレポジトリーの作出をおこない、生命科学諸分野で活用できる人類共通の財産を目指した。
(1)遺伝子試料の収集と細胞株の樹立:タイとインドネシアの集団について、細胞株化とDNA調製をおこなっている。現在1800株を得ている。
(2)汎用株化法のマニュアルを作成し、試験的に公開して使い勝手のモニタリングをした。
(3)供給体制の整備:集団を対象としたSNPのスクリーニング等に対応できるようDNA播種プレートの作成を試みた。
(4)汎用性・低コスト化の視点からの細胞株化技術の改良:EBウイルスを用いた株化では、時価1検体あたり200ドルのところを100ドル前後までコストダウンできた。T細胞株化法を2つ確立した。B及びTリンパ球同時株化法については、ドナー細胞の調整中である。
