| Research Abstract |
リウマチ性疾患との関連分析:東アジア3集団(日本、中国、タイ)において、全身性エリテマトーデス(SLE)とFc gamma receptor遺伝子群の比較関連分析を完了し、メタアナリシスを行った結果、FCGR2B-232T(P=0.0004)、FCGR3A-176F(P=0.0003)、FCGR3B-NA2(P=0.004)との関連を確認することができた。対照的に、米国白人SLE家族試料を用いたFc gamma receptor遺伝子群のTDT解析の結果、アジア集団とは異なり、FCGR2A-131R(P<0.005)との関連を見出した。多因子疾患における感受性遺伝子の集団差の例として興味深い。また、BLySに対する抑制型の受容体であるTACIの変異解析を施行し、腎炎陽性SLEとの関連、BLYS promoterとの組み合わせでSLE、関節リウマチ(RA)との関連を見出した。
疾患関連SNPの機能解析:SLEとの関連を示したFCGR2B-232Tアリルの産物について、FcgRIIb分子の膜ラフト分画への局在の減弱、およびBCR刺激による細胞内Ca^<2+>応答の亢進と持続時間の延長を見出した。これは、膜ラフトに局在する分子の機能的多型として最初の報告である。
病態関連遺伝子の機能解析:RA滑膜内血管内皮細胞における発現増強を見出したID遺伝子の役割を検討し、IDのみの強制発現でHUVEC(ヒト臍帯静脈血管内皮細胞)の増殖、活性化、血管新生を誘導することを見出した。
