Research Abstract

本研究では,扁桃体および海馬体を含む大脳辺縁系を中心とした情動・記憶・免疫システム間の相互干渉機構を,分子・遺伝子から細胞,個体(行動)レベルまでを含む学際的な研究チームにより総合的に解明した。
I.神経生理学的研究:若年者(20歳代)と中高年者(50歳代)を用いて、まず12種類の刺激音を4秒間隔で一定の順序で呈示し、音の呈示順序を記憶させた。ついで被験者に、記憶した順序と異なる順序で呈示された刺激音を識別させた(音順列記憶課題)。対照課題では、同じ12種類の刺激音をランダムの順序で呈示した。これらの課題に対する事象関連誘発電位(ERP)の解析により、300-700msecの潜時で、音順列記憶課題に特異的な陽性電位が頭頂-後頭部から記録されることが明らかになった。さらに、若年者と中高年者のERPおよび課題正答率の比較・解析により、i)若年者は中高年者より有意に課題正答率が高い、ii)若年者では、300-700msecの陽性電位が中高年者と比較して大きい、iii)課題の学習速度と陽性電位との間に、有意な相関が認められる、iv)これら陽性電位の電流発生源は、海馬体を中心とする側頭葉内側部に存在することなどが明らかになった。以上から、海馬体は順列記憶の形成に重要な役割を果していることが判明した。
II.神経解剖学的研究:ラットを用いて情動と記憶の相互作用に関与しているGABA系およびドパミン作動線維の連絡様式を解析した。その結果、ラット扁桃体基底外側核のGABAニューロンはドパミンD2受容体を発現し、ドパミン性入力線維とシナプスを形成していることが明らかになった。
III.分子生物学的研究:1)シナプス形成に何らかの役割を果たしている遺伝子Calminの発現をマウス上皮細胞を用いた解析により、calminがデスモゾームに存在することが示唆された。2)カイニン酸の腹腔内投与によるマウスの小脳性失調症に、小脳におけるIL-1ファミリー(IL-1β,IL-18)およびカスパーゼ1の活性化が関与することを明らかにした。3)遺伝子発現ベクター[弱毒βH1(HSV)ウイルス]に、神経保護作用のあるBDNF(脳由来神経栄養因子)遺伝子を挿入した組換えウイルスベクターを作製し、現在安定性を検討している。
IV.神経心理学的研究:統合失調症圏患者(統合失調症、統合失調型障害)を用いて、3D-MRIによる脳形態と探索眼球運動との関連をstatistical parametric mapping(SPM)により解析した。その結果、統合失調症に特徴的な探索眼球運動における反応的探索スコア(RSS)の低下と右下前頭回の灰白質の体積の減少が有意に相関することが判明した。
