Research Abstract	アルツハイマー病(AD)の発症には脳の老化が関与している。本研究では脳の老化により発現や機能が変化し、アルツハイマー病の発症に関連する因子を同定することを目的とした。 1.Neprilysin (Nep)はAβモノマーの主たる分解酵素で、脳の老化により発現が低下し、アルツハイマー病の発症促進因子となっている。そこで、Nepノックアウトマウスとプレセニリン1 (PS1)トランスジェニックマウスをかけ合わせ、AD病変の発現が促進するか調べる。現在、NEP^<-1->/PS1L286VとNEP^<-1->/PS1Wildのマウスが生まれ、最も古いもので8ヶ月齢となったので、脳の解析を開始した。 2.凝集Aβを結合するGlypican-1を強制発現させた細胞株を樹立した。これに凝集Aβ40をかけると細胞死が促進された。 3.Aβ1-42と結合する新規物質Adipは核移行シグナルを有し、Aβ存在下に細胞死を誘導した。 4.PSとcomplexを作る新規物質Adoplinに対するsiRNAを作用させると、Aβ産生が減少する傾向を見出したので、確認実験を行っている。
