| Research Abstract |
ヒト発癌のメカニズムを解明するため、横紋筋肉腫・胃腸管間質腫瘍より得られたDNAを使って、CGH(comparative genomic in situ hybridization)によるスクリーニングを行った。その結果、高度増幅領域が両者に高頻度に検出された。高度増幅領域は、横紋筋肉腫では1p31-p32、8q24、11q12-q13、12q13-14、18q12-q22、胃腸管間質腫瘍では3q25-28、5p、7q31であった。これらの領域で増幅している可能性のある既知の癌遺伝子を記載した;1p31-p32;c-jun、3q25-28;Evi1、PIK3CA、7q31;SLC2A2、met、8q24;c-myc、11q12-q13;bcl1、hst1、int2、12q13-14;CHOP、EWS、ATF1、WTF1、18q12-q22(q21);bcl2。横紋筋肉腫で見つかった1番と8番の高度増幅領域は、RT-PCR解析により1番染色体はc-jun、8番はc-mycであることが示唆される結果を得た。横紋筋肉腫で見つかった18番染色体長腕の高度増幅領域からは、染色体顕微切断・PCR法でペインティングプローブの作製に成功した。このプローブを使ってニホンザル、アカゲザルの染色体にFISHを行ったところ、特定染色体にバンド状の蛍光が見られ、サル染色体の同定に利用できることが分かった。胃腸管間質腫瘍で見つかった5番染色体短腕の高度増幅領域には癌遺伝子がほとんど報告されておらず、現在、解析を進めている。
