MNNGにより誘発したラット胃がんで異常なメチル化により不活化される遺伝子の同定

Research Abstract

N-Methyl-N'-nitro-N-nitrosoguanidine(MNNG)により誘発されたラット胃がんにおいて、過剰にメチル化されたDNA断片を、MS-RDA法によりゲノム網羅的に検索した。その結果、2個のクローンが分離され、11個の胃がんで、これら2クローンのメチル化の状態を検討したところ、11個全てで過剰にメチル化されていた。これらのクローンを含むBACクローンを分離、直接塩基配列決定法により、周囲1.4-3kbの塩基配列を決定した。1個のクローンから600bp離れたところにCpGアイランドが認められ、このCpGアイランドはgp115遺伝子の3'領域、Khk遺伝子の5'領域に相当した。これら2つの遺伝子の発現を、正常幽門腺及び胃がんで検討したところ、Khk遺伝子の発現には違いは認められなかったが、gp115遺伝子の発現は胃がんで特異的に誘導されていた。gp115遺伝子は、弾性線維の形成に重要な役割を果たすことが知られ、現在、gp115遺伝子を発現している細胞の種類、及び、メチル化異常と発現誘導の関連について、解析を進めている。
