| Research Abstract |
ヒト胃がん細胞株を材料に、methylation-sensitive-representational difference analysis法によるゲノム網羅的なメチル化異常の検索を行い、胃がん細胞株でメチル化された遺伝子5'領域のCpGアイランド9個を同定した。これら9個の下流遺伝子(LOX、HRASLS、FLNc、HAND1、Thrombomodulin、bA305P22.2.3、a homologue of RIKEN2210016F16、FLJ32130、PGAR)は、メチル化と相関して発現が消失していた。発現消失した細胞株を脱メチル化剤5-aza-2'-deoxycitidineにより処理すると、脱メチル化に伴う発現回復を認めた。胃がん臨床材料での検討では、前5個の遺伝子は、41例中12-17例(29%-41%)と高頻度にメチル化され、それに伴い発現が低下していた。後4個は、0-3例でメチル化されていた。LOX、HRASLS、Thrombomodulinについては細胞増殖抑制作用が報告されており、9個の中には胃がんに関与するがん抑制遺伝子が含まれている可能性がある。
