Research Abstract

ヒト正常型c-Ha-ras遺伝子トランスジェニックマウスは発がん物質投与により皮膚、肺腫瘍及び悪性リンパ腫を高率に発生することが知られているが、ヒトで高頻度にみられる乳腺、消化管、肝等には腫瘍の発生はみられない。一方、ラットでは化学発がん過程における前がん病変の発生からがんへの進展過程の解析や発がんを修飾する藷要因についてはマウスよりも遥かに多くの知見が得られているが、がん遺伝子トランスジェニックラットを用いた化学発がん研究は極めて少ない。本研究ではラットにヒト発がん関連遺伝子を導入し、化学発がん物質に対する感受性の亢進の有無を検索し、発がん感受性の亢進がみられた場合には導入遺伝子の果たす役割を明らかにし、さらに高発がん感受性モデルとして発がん物質の早期検索法の確立を目指した。
申請者が作成したトランスジェニックラットは、50日齢に乳腺発がん物質の投与でわずか8〜16週で全例に乳がんが多発し、乳腺発がん高感受性であることを明らかにした。またこのラットはニトロサミンによる膀胱発がん、および食道発がんにも高感受性であった。発生したがんではいずれも導入遺伝子に変異が認められ、発がん物質の標的となっていることが示された。乳腺発がんでは、高感受性の機序として、生後に新生乳管の形成を担う乳管先端部のterminal endbud(TEB)が、個体の成熟によってにおこる退縮が野生型より遥かに遅延し、野生型より多く存在していることを見いだした。従って発がん物質の標的組織と考えられているTEBの過剰残存が乳腺発がん高感受性の一因と考えられた。また、乳腺を標的としないニトロサミンの経口投与でも比較的短期に(雌10、雄20週以内)乳腺がんの発生に至ることから、これを指標とした環境発がん物質の中期スクリーニング法への応用の可能性が示された。
