| Research Abstract |
癌患者の免疫系が自己の癌細胞に発現するどのような分子を抗原として認識するのかを明らかにすること、またそれらの抗原中から癌の免疫療法のターゲットとなりうる癌抗原を同定することを目的として、研究を進めた。その方法として、癌組織由来の発現cDNAライブラリーを担癌患者のIgG抗体を用いて癌抗原を同定するSEREX発現クローニング法(Serological identification of antigen by recombinant cDNA expression cloning)を用いた。SEREX法は、癌患者の自家血清中の抗体が反応する癌抗原を同定する分子血清学的方法であり、患者の癌組織よりcDNA発現ライブラリーを作成し、患者血清中のIgG抗体と反応するクローンを単離し、cDNAの塩基配列を決定し抗原を同定する。SEREX法の利点は、培養細胞株の樹立を必要とせず、培養することが非常に難しい一般の固型癌からも癌抗原が同定できることである。また、樹立と保持が難しいCTLに依存する必要もなく、検出方法も簡便である。さらに、抗原の同定と遺伝子の同定が直結していることも大きな利点である。本研究の施行により、胃癌5症例から136種、乳癌2症例から76種、前立腺癌3症例から96種、肺癌1症例から63種、大腸癌1症例から35種の抗原を同定した。これらの中から診断・治療に有用な抗原を選択するために、(1)癌患者群と非癌患者コントロール群における各々の抗原に対する抗体産生頻度、(2)発現様式を検討することを行い、各々の癌で複数の有力な候補抗原を選び出した。
