| Research Abstract |
本研究では、「転写因子の活性制御機構及び、転写因子の異常によって生じる細胞機能の破綻を分子レベルで解明する」ことを目的とする。特に我々が同定したシグナル伝達因子TRAF6によって活性化される転写因子NFκBとAP-1の制御機構及びその異常によって誘導される細胞増殖・分化の破綻に焦点を絞る。以下の結果を得た。
1)昨年度同定したTRAF6結合分子TIFAは過剰発現系でIRAK-1と結合し、さらにTRAF6とIRAK-1の結合を顕著に促進した。一方、内在性TIFAはTRAF6と恒常的に結合しているのに対してIRAK-1とはIL-1刺激依存的に結合した。即ちTIFAはIL-1シグナルにおけるTRAF6とIRAK-1のリンカータンパク質であることが明らかとなった。2)TRAF6欠損マウスで見られる神経管閉管不全や胸腺の構築異常の原因を解明するためアフリカツメガエルを用いて初期胚発生におけるTRAF6とTIFAの発現を解析した。両遺伝子ともに神経胚初期の段階からmRNAの発現がはじまり、以降の段階で発現量が増加していた。Whole mount in situ hybridizationの結果、TRAF6は尾芽胚において咽頭弓、頭部の周辺部および脊髄で発現していた。咽頭弓は胸腺原基を形成する部位であるのでTRAF6欠損マウスにおける胸腺の構築異常の原因解明につながると考えられる。3)TRAF6の活性化と分解に関わるユビキチン化にTRAF6がE3ユビキチンリガーゼとして働くことを明らかにした。
