| Research Abstract |
受容体型チロシンキナーゼのリガンドからのparacrineによるシグナルとautocrineによるシグナルとの間に色素細胞の増殖・分化・ガン化に関する機能的な差異があるかどうかをin vivoで検証するため、HGFを環境側からparacrine的に供給するトランスジェニックマウスを作成した。このマウスでは、色素細胞の生存・増殖・分化が促進され、毛包囲外の皮膚に生涯色素細胞が定住し続けることが明らかになった。さらに、長期間の観察でも、メラノーマの発生頻度は野生型と有意な差はなかった。また、このマウスでは色素細胞がほとんど真皮に局在することが明らかになった。同じプロモーターを用いて皮膚に別の色素細胞増殖因子(SLF)を発現させた場合ほとんどの色素細胞が表皮に局在することを考えると、HGFは色素細胞が表皮へ移行するのに必要な分子の発現を抑制していると考えられる。われわれは、その分子の候補のひとつがE-カドヘリンであることを指摘した。新たに、HGFをautocrine的に供給するモデルとして、色素細胞前駆細胞から色素細胞特異的発現を誘導できるTRP-2遺伝子のプロモーターの下流にHGFを接続したDNAを用いてトランスジェニックマウスを作成した。樹立された7系統について経過を追っているが、少なくともへテロトランスジェニックマウスにはparacrineモデルでみられた色素細胞の増殖・分化の以上はみられなかった。
