Research Abstract	インテグリン活性化の初期段階においてはRhoAの活性は一過的に負に制御される。これは、細胞のleading edgeにおいては活性化されたRhoAによってストレスファイバーが形成されるのではなく、活性化されたCdc42やRac1によるmembrane ruffleを形成を促す為の機構であると解釈されている。我々は、インテグリン裏打ち蛋白質であるパキシリンが、インテグリン活性化に伴い4ケ所ののチロシンリン酸化を受けること、それら複数部位でのリン酸化の制御が上皮細胞運動におけるアクチンの再構成や細胞極性決定に重要な役割をなすことを明らかにし、報告してきた。今年度は、パキシリンの特定部位のリン酸化がRhoA活性の一過的負の制御に関与することを明らかにし、それに関わる分子群を同定しその分子機序を明らかにした。同じくインテグリン裏打ち蛋白質でありチロシンリン酸化されるp130CasはおそらくRac1を活性化しmembrane rufffle形成に寄与すると考えているが、我々の結果は、細胞形質膜におけるパキシリンのリン酸化制御によるRhoAの不活性化がmembrane rufffle形成に必須であること、従ってパキシリンとp130Casは互いに異なる分子機序によりmembrane rufffle形成に関与し、上皮細胞の運動を促すことを明らかにした。
