| Research Abstract |
悪性リンパ腫と胃癌をモデルとして、放射性同位元素(RI)標識モノクローナル抗体を用いるがんの特異的治療法開発に関する研究を行った。RIで標識した抗体は患者の体内で抗原・抗体反応に基づいて対応する抗原を発現しているがん細胞と特異的に結合するため,がんの画像診断としてのみならず副作用の少ない腫瘍特異的な治療法になると期待されている。本研究ではBリンパ球表面抗原CD20に対する抗体に細胞障害性の強い放射性ヨード(I-131)標識し研究に供したが、悪性リンパ腫細胞との強い結合を示した。倫理委員会の承認を得た後、B細胞悪性リンパ腫症例を対象としてI-131標識抗CD20抗体を投与したところ、ブドウ糖代謝を利用した核医学検査(FDG-PET検査と呼ばれる)では、脾臓のブドウ糖代謝の低下が認められ、治療効果が期待される.I-131標識ヒト型A33抗体による胃がんでの検討では、転移した胃癌細胞陽性リンパ節と転移のない腫大したリンパ節への抗体の集積の比率は最大26倍に達し、A-33抗体の胃癌への集積が認められた。またリポソームを使って癌細胞により多くのRI標識抗体を運搬する方法など基礎的研究も行った。
施設にRI治療病室が設置されたので、今後大量のI-131標識抗CD20抗体による悪性リンパ腫の本格的な治療が期待される。
