| Research Abstract |
mRNAによる免疫療法のin vivo効果を明らかにする目的で、実験モデルとしてB16メラノーマ移植マウスを用いて検討を行った。先ずmRNA単独の効果を見るために、mRNAを3回皮内投与後にB16細胞を皮下移植する予防実験と、B16移植後にmRNAを投与する治療実験を行った。mRNAとしてはEGFP(enhanced green fluorescence protein)、TRP2(tyrosinase-related protein 2)、TRP2-EGFPの3種を用い、これにPBSのみを投与する群を加えて計4群で検討した。PBS群に比較すると他の3群では腫瘍増殖の抑制傾向が認められたが、EGFP群とTRP2群との間では腫瘍増殖に差を認めなかった。次にmRNA導入樹状細胞(DC)を3回投与後にB16を皮下移植する予防実験を行った。この場合も4群の比較を行ったが、同様にPBS群と他の3群では後者で腫瘍増殖の抑制傾向が認められたが、EGFP群とTRP2群との間で腫瘍増殖に差を認めなかった。そこでB16の移植法を変更し尾静脈より注入する肺転移モデルでDCの効果を検討した。その結果、PBS群とEGFP群との間には差を認めなかったが、EGFP群とTRP2群の間で明らかな腫瘍増殖の差を認め、特異的免疫応答が惹起されたことが示唆された。さらにB16に発現するMAGE抗原群についても検討を進めている。これら動物実験のデータを基盤としてプロトコールを作成し、臨床試験に移行する予定である。
