| Research Abstract |
(1)昆虫細胞に遺伝子導入し、発現させたTL分子を用いてテトラマーを調製した。本TLテトラマーはTL抗原特異的細胞傷害性T細胞(CTL)をT細胞レセプター(TCR)/CD3依存性に染色し、本CTL群をin vitroで濃縮可能であることを示した。
(2)本テトラマーを用いて各種正常組織由来のTリンパ球を染色した所、脾臓や末梢リンパ節由来のCD8陽性リンパ球はほとんど染色されなかったが、予想に反してCD8陽性の腸上皮間リンパ球(iIEL)と胸腺細胞が染色された。この染色はCD8分子依存性で、TCR/CD3非依存性であり、その反応機序として、(i)CD8との直接反応、または(ii)CD8とTLリガンド分子の共役反応の2つの可能性が示唆された。
(3)TL抗原特異的CTL前駆細胞数をより正確に測定するために、CD8非結合性テトラマーの作成を試みた。CD8結合部位に変異を入れたTL分子(T3^b-A245V)を用い、テトラマーを作成し、その結果iIELや胸腺細胞とは反応しないTLテトラマーを得た。
