Research Abstract |
Objectives of Research After treatment of NaCl on a wheat line, cDNA fragments of expressed gene have isolated. The objectives of the research were; 1) to characterize salt-induced genes,2) to examine the genetic system of salt-tolerance/susceptibility in wheat, and 3) to isolate salt tolerant lines among wheat genetic resources, by utilizing the salt-induced genes. Summary of result: 1) By means of 5'RACE and sequence analysis, 5 novel ESTs (named as WEST genes) after salt treatment were,G6PDH gene, Mlo gene (known as powdery mildew resistance gene), a transformation factor gene, and two unknown ones. They showed expression peak within 12 hours after salt treatment, and their expression were relatively low in susceptible lines in comparison with tolerant lines of 6x wheat. 2) Chromosome locations of all 5 genes were determined. Also, QTL analysis was conducted on salt tolerance/susceptibility in a recombinant inbred population which revealed 8 significant loci in hexaploid wheat. Two of them were significantly linked with the locations of salt-induced genes. 3) Transform at wheat lines of a salt-induced gene, WESR4, with an enhanced promoter region were obtained, but they did not show clearly salt-tolerant in comparison with the non-treated.
