魚類のミトコンドリア・ゲノミクス:mtDNA全塩基配列決定に基づく解析

Research Abstract

核ゲノムに比べてサイズ的にもコンパクトなミトコンドリアゲノム(mtゲノム)は,動物の集団構造や系統のDNAレベルでの解析のための第1次的な分析対象として注目されている。しかし,魚類のmtゲノムに関する知見はまだ非常に限られているため,遺伝マーカーとしての有用性がまだ十分活かせない状況にある。そこで,1)魚類の主要なグループを代表する約30種について,mtゲノムの全塩基配列を決定し,2)その結果を詳細に比較分析することにより,魚類のミトコンドリアゲノムの実態と進化動態を明らかにすることを目的に本研究を開始した。われわれ自身が最近開発した効率的手法を活用した結果,今年度の目標分析種数(30種)のmtゲノムの全塩基配列を決定することができた。とくに今年度は,現生魚類の大部分を占め,かつ有用水産資源の過半を占める真骨類を中心に取り上げたが,中でも"低位"魚類に力点をおいて研究を進めた。また,得られた塩基配列データの解析を行っているが,脊椎動物で初めて見出された遺伝子配置の変動がいくつか見つかった。さらに,塩基配列データを基に,予備的な分子系統学的解析を行った結果,ウナギなどが属するカライワシ類が従来想定されていたよりも系統的により"低位"に位置することなど,新たな知見が得られた。以上のように,魚類のミトコンドリアゲノムの実態と進化動態の解明に資する情報の蓄積が着実に進んでいる。
